چکیده
introduction: interleukin-2 (il-2) as a t helper type 1 (th1) cytokine has an important role background and objective: interleukin-2 (il-2) as a t helper type 1 (th1) cytokine has an important role in activation, growth and differentiation of several immune cells. moreover il-2 is known as a pro-inflammatory and anti-tumoral cytokine. in addition deysregulation of il-2 in some diseases such as autoimmunity has been shown. the present study was conducted to evaluate the patterns of il-2 production in human peripheral blood mononuclear cells (pbmcs) and human leukemic t cells lines (molt-4 and jurkat). materials and methods: human pbmcs and leukemic t cells were cultured in complete rpmi-1640 medium. the cells were then seeded at a density of 106 cells/ml and were incubated with different concentrations of phorbol myristate acetate (pma) (1-25 ng/ml) or phytoheamagglutinin (pha) (2-10 μg/ml) for 48 hours. then the cell-conditioned media were collected and used for il-2 assay. statistical comparisons between groups were made by analysis of variance (anova). results: pha/pma significantly and dose-dependently increased il-2 level in human pbmcs and leukemic t cells (molt-4 and jurkat) after 48 hours of incubation compared with untreated control cells. conclusion: we conclude that human pbmcs and leukemic t cells (jurkat and molt-4) could potentially secrete il-2 with different degrees. thus, these cells could offer a proper system to study the mechanisms regulating il-2 production in diseases in which il-2 production is disregulated. also these cells could be useful for screening the il-2 modulators.

متن کاملEvaluation of in vitro Effect of Pyocyanine Pigment on Interleukin-2 Secretion from Peripheral Blood Mononuclear Cells in Cancer Patients
Background and Objective: The pyocyanine pigment in Pseudomonas aeruginosa stimulates blood cells to secrete IL-2. IL-2 cytokine is an activator of cytotoxic T cells and natural killer cells. These cells destroy the target cells of patients with cancer. This study aimed to evaluate the effects of pyocyanine on the IL-2 secretion from peripheral blood mononuclear cells (PBMCs) in patients with b...
متن کاملPancreatic Cancer Cell Lines Can Induce Prostaglandin E2 Production from Human Blood Mononuclear Cells
Accumulating evidence suggests an important role for cyclooxygenase-2 (COX-2) in the pathogenesis of a wide range of malignancies. The protumorigenic properties of COX-2 are generally thought to be mediated by its product, PGE(2), which is shown to promote tumor spread and growth by multiple mechanisms but most importantly through modulation of the local immune response in the tumor. Pancreatic...
متن کاملInterleukin 2 inhibits in vitro growth of human T cell lines carrying retrovirus
Four human T cell lines, TL-Mor, TL-Su, TL-TerI, and TL-OmI, carrying human T cell leukemia virus (HTLV), were established previously. TL-Mor, TL-Su, and TL-TerI were derived from interleukin 2 (IL-2)-dependent parental cell lines cloned from peripheral blood leukocytes (PBL) of three healthy HTLV carriers, while TL-OmI was directly established from PBL of a patient with adult T cell leukemia. ...
متن کاملProduction and regulation of interleukin-2 in human lymphoblastic leukemias studied with T-cell monoclonal antibodies.
Human leukemias are illnesses of hemopoietic stem cells that go through processes of self-replication and partial differentiation under the control of as yet largely unknown growth and differentiation factors. IL-2 is a powerful factor controlling proliferation of normal T cells. We report that acute lymphoblastic leukemias of T and non-B, non-T phenotypes produce a growth factor after mitogen ...
متن کاملInterleukin-17A and Interleukin-17F mRNA Expressions in Peripheral Blood Mononuclear Cells of Patients with Multiple Sclerosis
Bakground: Multiple sclerosis (MS) is a CD4+ T cell-mediated autoimmune disease affecting the central nervous system (CNS). It was previously believed that Th1 cells were pathogenic T cells in experimental autoimmune encephalomyelitis (EAE). However, the functional role of Th1 cells in EAE has been reconsidered upon the discovery of IL-17- producing T cells which are consider as dominant effect...
